Formation 5'-ethylthioadenosine from DL-ethionine in yeast.

چکیده
Experiments on the formation of adenine thiomethylriboside from methionine in yeast (1) suggested a study of ethionine as a source of organic sulfur in the growth medium. The sulfur atom of this amino acid analogue is not utilized for the formation of thiomethyladenosine (2). However, we found in extracts of yeast (Torulopsis utilis and Saccharomyces cerevisiae) grown in the presence of an excess of nL-ethionine another nucleoside which seemed to be closely related to thiomethyladenosine. Besides showing the typical ultraviolet absorption spectrum of an adenine compound, it contained sulfur and gave a nitroprusside reaction like that of methionine, ethionine, or thiomethyladenosine (3). The differences in the RF values on paper chromatograms precluded the possibility that the new compound was identical with thiomethyladenosine. It appeared probable that ethylthioadenosine could be formed when ethionine was present in the growth medium. The isolation of this substance in quantity and its analytical examination corroborated this assumption.

Metabolism of ethionine in ethionine-sensitive and ethionine-resistant cells of the enteric yeast Candida slooffii.
In a defined medium with added ethionine plus low methionine, phenylalanine, tryptophan, tyrosine, adenine, and additional methionine reversed inhibition of the enteric yeast Candida slooffii by ethionine. Isoleucine and 7-methylguanine restored half-maximal growth. Choline but not triethylcholine inhibited C. slooffii. 6-Mercaptopurine reversed ethionine inhibition and also synergistic inhibit...

Since the original report on ethionine (I) by Dyer (I), numerous studies have demonstrated the inhibitory properties of this analogue of methionine on growth in higher animals and microorganisms (cf. Woolley (a)), and even on the propagation of viruses (3, 4) and the reversal of these inhibitions by methionine. A number of pathological effects of ethionine on different organs have also been rep...

متن کاملFormation of S-adenosylethionine in liver of rats chronically fed with DL-ethionine.
The early changes in the metabolism of L-ethionine were examined in rats preexposed to chronic administration of DL-ethionine. The capacity of liver to accumulate S-adenosylethionine after a single injection of L-ethionine decreases rapidly from the onset of the carcinogenic regimen. This drop is caused by diminished S-adenosylethionine synthesis, a consequence of lower activity of the ATP-L-me...
متن کاملAflatoxin biosynthesis in Aspergillus parasiticus: effect of methionine analogs.
The physiological effects of DL-ethionine and related methionine analogs upon cellular protein synthesis and methyl group transfer to aflatoxin B1 in Aspergillus parasiticus were examined in growing and resting cell systems. The addition of DL-ethionine (0.02 M) to growing mycelia inhibits cellular growth some 37% after 120 h of development. Addition of ethionine before 50 h prevents any aflato...
